本試驗測定紅細胞對各種濃度低滲溶液的抵抗力。在低滲鹽水中，水分透過細胞膜，使紅細胞逐漸脹大破壞。紅細胞的滲透性主要取決于紅細胞的表面積與體積之比。表面積大而體積小者對滲鹽溶液的抵抗力較大，反之則抵抗力較小（脆性增加）。球形紅細胞表面積/體積比值減少，對低滲溶液特別敏感，脆性顯著增加。
